Abstract Aniline degrading bacteritum, Flavimonas oryzuhabitans and 2,4-D degrading bacterium, Alcaligenes sp. were fused by spheroplast fusion in order to develop the news strains of microorganisms having the degradative ability for various aromatic hydrocarbons, and the characteristics of the obtained fusants were analyzed. The frequency of recombinant formation was 1.01×10^-6, and 0.4% of the obtained recombinants, which were selected in minimal regeneration media, showed the degradative ability in minimal liquid selective medium added aniline or 2,4-D respectively as a sole carbon source. Aniline degfadation by fusant strain(FA3) was increased about 5.2-fold to compares with that of the parental strains, while the degradative ability of 2,4-D was slightly lower than that of parental strains. Genetic relatedness between a fusant(FA3) and the parents were estimated by DNA-DNA hybridization with Dig-labelled chromosomal DNA fragments of F. oryzihabitans and Alcaligenes sp. as prove respectively. DNA of the fusant FA3 showed a cross-hybridization with both probes, indicating a DNA homology between the fusant and both parental strains. The G+C ratio of F. oryzihabitans, Alcaligenes sp., and FA3 were 43.04±3.98, 56.97±3.98, and 62.84±3.98, respectively.
