Abstract |
The shuttle vector pYWH02 that is able to replicate in A. tumefaciens and E. coli was constructed using the pBR322 and a cryptic plasmid pTi12 isolated from A. tumefaciensKU12. The size of pYWH02 is 11.56 kb and 7.2kb HindIII fragment of pYWH02 contains the replication origin of pTi12. In order to reduce the size of shuttle vector, subcloning was performed. As s result of subcloning, 4.69 kb pYWEE was contructed using the pUC19 and 2kb EcoRI-EcoRI fragment including the replication origin that derived from the 7.2kb HindIII fragment of pTi12. |
