| Abstract |
Salmonella schottmulleri의 배양액에서 황산암모늄 침전법, DEAE Sepharose Fast Flow ion exchange chromatography 및 Sephacryl S-200 HR molecular sieve chromatography를 이용하여 cytoxiv protein을 정제하였다. 정제된 cytotoxic protein의 분자량은 SDS-PAGE상에서 약 55kDa이었고, molecular sieve chromatography에 의해 약 120kDa으로 나타나 정제된 cytotoxic protein은 동일 단위체로 구성된 이합체로 생각되며, 등전점은 6.6이었다. 정제된 cytotoxic protein은 papain, pepsin, proteinase K, trypsin 등에 의해 분해되어 세포독성 활성이 소실되었으나 lysozyme에 의해 분해되지 않고 세포독성 활성을 유지하였다. 최대 세포독성을 보인 cytotoxic protein의 최소량은 30㎍/㎖이었으며, 정제된 cytotoxic protein은 열에 불안정하였다. |
